Single Cell Genomics

The Core Immunology Lab, in collaboration with the Gladstone Genomics Core and the UCSF Center for Advanced Technology is offering a service pipeline for single cell genomics of samples up to BSL2* (BSL2 with enhanced precautions) using the 10x Genomics platform.

This pipeline consists of the following:

UCSF Core Immunology Lab

  1. Sample preparation and QC (Core Immunology Lab)
  • Thawing of frozen samples (if necessary)
  • Removal of dead cells by magnetic selection
  • Nucleated cell counting and viability assessment using a volumetric flow cytometer
  • Sample multiplexing (if desired)
  1. Single cell GEM generation using the 10x Single Cell Controller
  2. Sample cleanup, RT, and cDNA preparation
  3. Courier to the Gladstone Genomics Core

Gladstone Genomics Core

  1. Sequencing library preparation and QC
  • Bioanalyzer of cDNA
  • Library prep
  • Bioanalyzer final libraries
  • qPCR of final libraries
  • Pooling of samples for sequencing
  • Sample submission for sequencing at CAT or at the Gladstone Genomics Core
