The Core Immunology Lab, in collaboration with the Gladstone Genomics Core and the UCSF Center for Advanced Technology is offering a service pipeline for single cell genomics of samples up to BSL2* (BSL2 with enhanced precautions) using the 10x Genomics platform.
Please contact us at [email protected] to provide us information regarding your project.
This pipeline consists of the following:
UCSF Core Immunology Lab
- Sample preparation and QC (Core Immunology Lab)
- Thawing of frozen samples (if necessary)
- Removal of dead cells by magnetic selection
- Nucleated cell counting and viability assessment using a volumetric flow cytometer
- Sample multiplexing (if desired)
- Single cell GEM generation using the 10x Single Cell Controller
- Sample cleanup, RT, and cDNA preparation
- Courier to the Gladstone Genomics Core
- Sequencing library preparation and QC
- Bioanalyzer of cDNA
- Library prep
- Bioanalyzer final libraries
- qPCR of final libraries
- Pooling of samples for sequencing
- Sample submission for sequencing at CAT or at the Gladstone Genomics Core
